Spravato’s mechanism of action is unlike traditional medications. Whereas most oral antidepressants work on the serotonin receptor (SSRIs, for example) and may have many negative side effects, Spravato works through a different pathway. Spravato (esketamine) is an NMDA receptor antagonist, which means it works by modulating the activity of the neurotransmitter glutamate. Glutamate is involved in the communication between nerve cells and is associated with the formation of new neural connections and pathways.
